Courtesy of Professor Frederick Sachs at SUNY - There are three commons sources of free energy to drive physiological processes: chemical potential, electrical potential and mechanical potential. The first two have been the subject of many optical probes, but measuring the stress in living cells has not. We have developed FRET based probes that measure the mechanical stress in specific cytoskeletal protein in real time. We have been able to measure stress induced changes in the probe itself in solution using DNA springs; internal stresses in proteins such as filamin, spectrin, in living cells, and intrinsic stress in collagen in living worms. We are making a transgenic mice with specific proteins labelled with the stress probes.
